The Trypsin-EDTA solution contains 0.25% Porcine Trypsin and 0.1% of EDTA in in Hanks’ Balanced Salt Solution with Phenol Red. Trypsin is a member of the serine protease family. It consists of a single-chain polypeptide of 223 amino acid residues. Ethylenediaminetetraacetic acid (EDTA) is a chelating agent and added to enhance enzymatic activity of trypsin solution. The product is suitable for mammalian cells such as adherent and suspension cell lines.

Application

Dissociation of cells

Remove the medium from the flask/vessel by aspiration. Wash the cells once with 1x Dulbecco Phosphate buffered saline without calcium and magnesium for 2 minutes. Discard the DPBS, add Trypsin-EDTA solution and incubate in incubator for 1-2 minute or until the cells are dislodged from the surface. Monitor the process by observing the flask under inverted microscope. Once the cell dissociation is complete, Flush the cells with complete Media to the flask to inhibit the tryptic activity. Count and sub-culture the cells as required. Concentration of Trypsin-EDTA to be determined by the end user.

Storage

Store at -20°C for long term, avoid repeated freeze thaw, Aliquots can be stored at -20° C for regular use; The Product must be used within the expiry given on the product label.
