The principle of combining liquid phase chip and double antibody sandwich is used. Two microspheres of different sizes (4μm and 5μm) with different fluorescence intensities are coated with antibodies specific to the detection index (capture microspheres), which can be specifically bound to the corresponding antigen proteins in the sample, and biotin-labeled detection antibodies are added to form an immune complex of antibody capture microspheres-cytokines-detection antibodies. Wash to remove excess unbound antibodies, add streptavidin-phycoerythrin conjugate (SA-PE), and detect the corresponding fluorescence on flow cytometry. The fluorescence intensity of the immune complex is used to obtain the content of the sample index to be tested.
