ATP is the main energy molecule in cells and closely reflects metabolic activity. Since ATP levels are proportional to the number of viable cells, measuring ATP provides a reliable way to assess cell viability.
This kit uses Firefly luciferase, which catalyzes the reaction of luciferin with ATP and O₂ in the presence of Mg²⁺, producing light. The luminescence intensity is directly proportional to ATP concentration, and therefore to the number of viable cells.
The assay is highly sensitive, stable, and easy to use. In a 96-well format, it shows a strong linear response from 5 to 100,000 cells per well (the upper limit may vary by cell type). The procedure is fast—results are obtained in about 10 minutes—with no need for washing or medium replacement. The reagent has a long half-life and excellent stability. Compared with Calcein-AM and CCK-8 assays, the ATP-based method is simpler, faster, and more efficient.
