The HyPure Universal DNA Kit is designed for purification of genomic DNA from a wide range of biological samples using silica membrane spin column technology. The optimized extraction chemistry supports efficient DNA recovery from complex sample matrices including blood, tissue, buffy coat, body fluids, swabs, dried blood spots and sperm samples.

The kit combines Proteinase K digestion with silica membrane adsorption to efficiently release genomic DNA from biological samples while removing proteins, lipids and other contaminants. The resulting DNA is suitable for routine molecular biology workflows such as PCR amplification, genotyping analysis and HLA typing studies.

Workflow Overview

The HyPure Universal DNA Kit uses a silica column-based workflow for purification of total DNA from routine sample types including tissue, blood, body fluids, cultured cells and swab-based materials. After sample-specific pretreatment, DNA is released by protease-assisted lysis, captured on the silica membrane under ethanol-containing binding conditions, and recovered through washing, drying and elution.

Sample Handling Logic

The main variation between sample types lies in the pretreatment and digestion stage. Tissue samples require cutting and complete digestion, cultured cells require pellet resuspension before lysis, while blood and other liquid samples can usually enter lysis more directly. Once the binding conditions are established, the downstream purification steps remain largely consistent.

Time and Workflow Characteristics

Under typical manual operation, the overall workflow for routine samples is usually completed within about 30–90 minutes, depending mainly on sample pretreatment and digestion requirements.

Proteinase K should be stored at 2-8°C upon arrival. However, short-term storage (up to 12 weeks) at room temperature (15-25°C) does not affect their performance. The remaining kit components can be stored at room temperature (15-25°C) and are stable for at least 18 months under these conditions. The entire kit can be stored at 2-8°C, but in this case buffers should be redissolved before use. Make sure that all buffers are at room temperature when used.
