| Description | Dulbecco’s Modified Eagle Medium-Low Glucose (DMEM) Powder
With 1gm Glucose per liter, L-Glutamine and Sodium pyruvate Without Sodium bicarbonate Dulbecco’s Modified Eagle Medium (DMEM) is a widely used cell culture medium derived from Eagle’s medium. It is an enriched version of Basal Medium Eagle (BME), containing four times the concentration of amino acids and vitamins. DMEM also includes glycine, serine, and ferric nitrate. The original formulation, developed for culturing embryonic mouse cells, contains 1000 mg/L of glucose. |
| Note | It is recommended to monitoring the pH of the medium. If needed, pH can be adjusted by using sterile 1N HCl or 1N NaOH. |
| Directions for Use | 1.Suspend the pouch content in 900 ml of ultra-pure water or water for injection grade suitable for tissue culture with constant stirring until the powder is completely dissolved.
2.Weigh 3.7 g of sodium bicarbonate powder and add to 1 litre of medium and stir until dissolved. 3.Adjust the pH to ~0.25 below the desired pH using 1N HCl or 1N NaOH, since the pH tends to rise during filtration process. 4.Make up the volume to 1000 ml with water. 5.Sterilize filter through a sterile membrane filter with a porosity of 0.22µm, using positive pressure to avoid the loss of carbon dioxide. 6.Aseptically add sterile supplements as required and aliquot sterile medium into sterile cell culture grade bottles. 7.Store liquid medium at 2-8°C till further use. |
