Cultural Response Incubate the tubes (with caps tightened) at 35 ± 2°C for 18-48 hours or upto 96 hours before interpreting the results as negative. After the incubation period, observe the tubes for growth and decarboxylase reaction. In the initial stages of incubation the medium colour changes from purple to yellow due to fermentation of glucose by the organisms, with acid production. On further incubation, L-Lysine is decarboxylated to cadaverine, there will be an alkaline reaction (rise in pH) and the colour of the medium changes to purple. If colour remains yellow, the decarboxylase reaction is negative.
