| Principle | Detection Principle of Pituitary adenylate cyclase activating polypeptide(PACAP) ELISA Kit As mentioned above, this kit utilizes the Double Antibody Sandwich ELISA technique. The pre-coated antibody is an anti-Pituitary adenylate cyclase activating polypeptide(PACAP) monoclonal antibody, while the detection antibody is a biotinylated polyclonal antibody. Samples and biotinylated antibodies are added to the ELISA plate wells, and each addition is followed by washing with PBS or TBS. Then avidin-peroxidase conjugates are added to the wells. After thoroughly washing away the unbound enzyme conjugates with PBS or TBS, TMB substrate is added for color development. TMB is catalyzed by peroxidase to form a blue product, which finally turns yellow after the addition of stop solution (Color Reagent C). The color intensity is positively correlated with the concentration of the target analyte in the sample. |
